Usage 1. Dilution of fluorescent dye: If the dye is dry powder, use 100ul DMSO to dissolve it to make 100X dye mother liquor; If the dye is liquid, it means that it is 100X mother liquor dissolved in DMSO from the factory; Use the signal amplification reaction solution to dilute at 1: 100 to prepare the dye working solution (ready-to-use); DAPI prepares the working solution using sterile water dilution at 1: 100. 2. Use of secondary antibody: This kit is matched with a universal secondary antibody for pika and rabbits, which is not suitable for mouse tissues. Please verify whether the species of the primary antibody matches before the experiment. Please refer to (# abs50030) for mouse tissue samples.

Theory Multiplex fluorescence immunohistochemical staining is based on the principle of specific binding of antigen and antibody. The secondary antibody labeled with horseradish peroxidase (HRP) is selected to activate the fluorescent dye in the kit and covalently bind the signal to the antigen. In situ multi-target staining of tissues or cells is achieved by multiple rounds of staining cycles with the aid of different fluorescent dye labels.

Description There are complex cells in tissue microenvironment, and the phenotype, state, abundance and distribution of these cells have important biological significance and clinical value. It can be rendered in situ in tissue by means of antibody staining. Immunohistochemical staining is a common technique to study tissue morphology and protein expression in situ. Routine IHC detection can only show a single indicator, and it is difficult to present the cell composition, state and relationship in the complex tissue microenvironment. This information is crucial to the diagnosis and treatment of diseases!
Principle of tyrosine signal amplification technology: Similar to the DAB color development method of conventional immunohistochemistry, TSA technology also uses HRP-labeled secondary antibody. HRP catalyzes the fluorescein substrate added to the system to produce activated fluorescein substrate, which can be covalently bound to the antigen. Tyrosine is covalently bound to stably covalently bound to fluorescein on the sample. After that, the non-covalently bound antibody is washed away by thermal repair method, and then the next primary antibody is replaced for the second round of incubation, and another fluorescein substrate is replaced, so that multiple labeling can be achieved by reciprocating.

General Notes 1. This kit is only used for immunohistochemistry and is not used for other purposes. 2. This kit is restricted to professional use only. 3. Appropriate protective measures should be taken to avoid contact of the reagent with the skin and eyes. 4. The activity of reagents beyond the expiration date may be reduced, so reagents beyond the expiration date should not be used. 5. If the dyeing components of this kit are mixed with products of other companies, abnormalities may occur during the dyeing process. 6. Incomplete dewaxing will easily affect the dyeing effect. 7. In order to prevent possible false negative and false positive results, positive control and negative control should be carried out at the same time during the experiment. 8. All kinds of wastes generated in the use of this kit should be disposed of in accordance with the Regulations on Medical Waste Management.
Storage Temp. Fluorescent dyes should be stored in the dark from light at 2 ~ 8 ℃, and the validity period is 12 months from receipt of the goods.
Applications It is mainly used for immunohistochemical staining of tissues, paraffin sections and TMA chips. It can also be used for frozen sections and cell crawling sections, and needs to be matched with abs994 antibody eluate (specific for mIHC).

The texture is great, had smooth areas and rigid areas all while being soft and durable. The product is pretty thick, which I needed for my Pit-Bull. This product however lasted years before I started to notice damage. It is one of her favorite toys. She liked it so much I bought two, one for the house and one for the car for road trips. It’s the perfect toy to keep a chewer distracted inside a car, along side a deer antler, which also helps with dental hygiene. She gets dental hygiene compliments every trip to the Vet. We use multiple different things to help with her teeth and breathe but, I think this toy contributes the most to her healthy teeth. I call it a toy because she plays with it all the time. My dog loves it, and me and my house are soo glad he's chewing on this! If you have a super chewer (I'd never heard of that) I strongly suggest this toy. They also have larger ones for bigger breeds!

Why did you pick this product vs others?: I bought the Pawbler because the two most popular wobble treat feeders on the market are plastic, and one of my dogs just absolutely destroys those in no time because he bites them and throws them rather then nudging them and then he has plastic cutting him so I am forced to throw away a costly treat toy/dispenser. I thought maybe a rubber one would work out better. So far my dogs love it, it works well spitting out their kibble, seems to be holding up and is easy to clean and feels like a good weight, not a super flimsy toy. I would love to see it made in a slightly larger size, I think this current size is good for medium dogs. It holds just under a cup of small round kibble but I would like to see one that can hold more and is slightly larger in size. I have 4 german shorthaired pointers, my test subject for this toy is mostly my 70 pounder who bites the toys instead of nudging them and throws them across the room, so it is taking a beating. I think for him a larger size would be helpful in deterring that behavior but I also think this may be too small for larger breeds. Durability: So far this toy looks just like when I bought it. It has no punctures or scratches and unscrews without problem. We have only had it a week or so but we use puzzle/toy feeders to feed our dogs lunch every day so it is pretty easy to tell which toys won't last long. Obviously, I can't speak to long-term durability at the moment, but I can say that most toys similar to this end up already scratched or punctured in a few days, so this is already sturdier when compared to what I have tried before.
